b. Vectors for Agrobacterium-mediated method


The vector/Agrobacterium strain combination that have been performing consistently in our facility is pPZP derived vector1 and EHA1012. We strongly recommend to use this vector/strain combining with the bar gene/bialaphos selection for maize and soybean services in our facility. For rice and Brachypodium, both bialaphos and hygromycin can be used for selection. One recommended binary vector is pTF101.13 (see below for detail)

We can also perform transformation services using other vector/strain/selection systems upon customer request. However, such proposal needs to be thoroughly discussed with the facility (contact kanwang@iastate.edu) as a non-service collaboration and no transgenic product can be guaranteed.
