Rice Genetic Transformation:
Agrobacterium-mediated

Nippon Bare rice seed - Starting material for callus initiation and transformation

The PTF Rice Transformation System:

Target tissue: Callus culture derived from mature embryo
Transformable cultivars: Japonica cv. Nipponbare Delivery means: Agrobacterium tumefaciens
Selection: the bar gene/bialaphos or the hpt gene/hygromycin
Products available: Plantlets, seed
Protocol: please see Rice Transformation Protocol

Transformation frequency:

25%

Transformation frequency is defined as the number of bialaphos resistant callus per 100 infected callus pieces.

To order your transgenic rice:

  1. Obtain APHIS permit
  2. Request Agrobacterium vector
  3. Submit your construct
  4. Receive transgenic plantlets

  5. OR
    Receive transgenic R1 seeds

Time frame from infection to transgenic seed:

Transformation Activity Day
Initiate callus from mature seed 1
Select callus for transformation 35
Transform callus tissue 39
Pick and bulk bialaphos resistant callus events 88-102
Regenerate transgenic plantlets 109-123
Harvest transgenic seed 220
