Maize Genetic Transformation:
Agrobacterium-mediated

Bialaphos resistant event regenerating to produce plants

PTF Maize Transformation System:

Method optimized in PTF lab (Frame et al. 2002)
Target tissue: Immature zygotic embryo
Transformable genotypes: Hi Type II hybrid
Delivery means: Agrobacterium tumefaciens
Selection: the bar gene/bialaphos
Products Available: Plantlets, seed, callus
Protocol: please see Maize Agrobacterium Transformation Protocol

Transformation frequency:

7%

Transformation frequency is defined as the number of bialaphos resistant callus per 100 infected embryos.

To order your transgenic maize:

  1. Obtain APHIS permit
  2. Request Agrobacterium vector
  3. Submit your construct
  4. Receive transgenic plantlets
    OR
    Receive transgenic seeds

Time frame from infection to transgenic plant/seed:

Transformation Activity Day
Embryo infection 1
Pick and bulk bialaphos resistant callus events 60-90
Regenerate transgenic plantlets to soil 80-110
Harvest transgenic seed in greenhouse 190-220
