Establish transgenic analysis system


The choice of which transformation event to regenerate is the customer’s responsibility. Approximately 12 weeks after submitting your construct, you will be notified by e-mail that you will be receiving a large number of transgenic callus samples that will need to be processed within 10 working days. The samples will be shipped via UPS or FedEx overnight service.


You should expect about 60-90% of the samples that were co-bombarded with our plasmid containing the Bar selectable marker to also carry your gene of interest. Once you have selected the events you want regenerated (to plants), please fax the confirmation sheet to us.


Depending on your experiment, the parameters for choosing an analysis system may vary. To help you make this choice, we recommend using one of the following protocols:


  1. Callus DNA extraction
  2. Callus RNA extraction
  3. Callus protein extraction
  4. PCR
  5. Southern blot analysis
  6. Northern blot analysis
  7. Western analysis
  8. ELISA

NOTES:The efficiency of analysis protocols may vary depending on the parameters used for each different transgene analyzed. The present protocols have been set up for Bar detection.


A rapid screening can be done by PCR (or RT-PCR for transgene expression). Those protocols vary depending on transgene.


The protocols listed here have been used in our laboratory successfully but by no means are the only protocols for molecular analysis. The efficiency of analysis protocols may vary depending on the parameters used for each different transgene to alalyze. The present protocals have been set up for Bar detection.
